What if this immune function also exists in humans, but hadn’t been discovered yet?
We analyzed SIR2 from thousands of bacterial, archaeal & eukaryotic genomes. All housekeeping sirtuins group in a clade, whereas all antiphage SIR2 group in another one that we named SIRim, for “SIR” and “immunity”.

Akin to bacterial SIR2 antiphage proteins, human SIRal, also known as FAM118b, forms filaments that are essential for its NAD processing activity.

Awesome to see structures of these filaments that differ from bacterial ones.

Many components of human immunity evolved from ancient pathways in bacteria. A beautiful story from @audeber.bsky.social‬ @enzopoirier.bsky.social‬ leverages this evolutionary connection to discover SIRal as a signaling protein in animal TLR-mediated innate immunity.

I am so happy to share this new paper I co-wrote with colleagues at the USGS. It's a guide to predicting how nonmotorized recreation like hiking, skiing, mountain biking, etc. affects ungulates (hoofed mammals like deer, elk, moose, and pronghorn) to inform NEPA analyses. doi.org/10.3133/sir2...

Could the knowledge of bacterial immunity help us discover new human immune proteins?

One protein family caught our attention: the SIR2 proteins. In humans, they are called sirtuins and notably known to regulate gene expression — but in some bacteria, they defend against viruses.

Skipping some stuff, we found that Sir2 is a key regulator for this structure, as the rDNA loop collapses when Sir2 is gone. We also found that Sir2 is required for proper quiescence entry and exit. So many roles for Sir2 out there!

We started thinking about chromatin structure at the nucleosome level (can't help ourselves) & saw that in cells lacking Sir2, chromatin is more repressive based on H3 ChIP-seq. Pol II however was increased across the locus, even in quiescence, when Sir2 was gone.

SIRim proteins are conserved across highly diverse phyla from bacteria to fish, fungi and protists.🐟🍄🐘

Among this large family of proteins, present in thousands of living organisms, we identified on human protein, which function was previously unknown.

We named it SIRal, for SIR2 antiphage-like.
