Pharmacological inhibition of METTL1 as a therapy against solid and blood cancers
Sf21 cells in Sf-900 II serum-free medium (Gibco, 11497-013) were co-infected at a multiplicity of infection of 1 for both His-tagged METTL1 and Flag-tagged WDR4 baculovirus preparations. The infected cultures were incubated for 72 h at 110 rpm and 27 °C. Cells were collected by centrifugation at 3,400g for 15 min at 4 °C and the pellets were stored at −80 °C. The pellets were lysed in 25 mM Tris pH 8.0 (Invitrogen, 15568025), 300 mM NaCl (Merck, S6546), 1 mM tris(2-carboxyethyl)phosphine (TCEP) (Thermo Fisher Scientific, 20490), 5% glycerol (Thermo Fisher Scientific, 327255000), 0.25% CHAPS (Thermo Fisher Scientific, 28300), 1 mM phenylmethylsulfonyl fluoride (PMSF) (Thermo Fisher Scientific, 36978) supplemented with protease inhibitor tablets (Roche, 4693124001) and 10 U ml−1 benzonase (Merck, E8263). Homogenization was achieved by two cycles at 20 kilopounds per square inch using an IKA high pressure homogenizer. Lysates were briefly centrifuged in between cycles (2,000 rpm for 15 min) to remove foam and finally cleared by ultracentrifugation (45 min at 150,000g, 4 °C). The protein complex was purified sequentially by immobilized metal affinity chromatography (IMAC), desalting, ion-exchange chromatography (IEX) and size-exclusion chromatography (SEC). IMAC purification was performed on the 5 ml HisTrap FF Crude column (Cytiva) at...

I am really, really irrationally annoyed by our chemical inventory system claiming an entry was "AI generated" when I have to enter all of the data manually. And sometimes it overwrites my SMILES code and removes the structure entirely.

Toxic flame retardants behave differently in saltwater versus freshwater food webs, study finds: TCEP biomagnifies in rivers while diluting in oceans
Organophosphate esters (OPEs) are used in furniture, electronics, textiles and building materials. Because they are physically mixed in rather than chemically bound, they leach out continuously into air, dust and water. Once in aquatic environments, they are taken up by algae, ingested by small animals and passed up the food web. Some OPEs are known endocrine disruptors, neurotoxicants and carcinogens. Most research has focused on either marine or freshwater systems separately, leaving a critical gap: no one knew whether a chemical that dilutes in the ocean might magnify in a river – or the reverse. Given these uncertainties, a systematic cross‑ecosystem comparison became essential. A team from Changchun Normal University and the Chinese Academy of Sciences has conducted the first side‑by‑side comparison of OPE bioaccumulation and trophic transfer in contrasting aquatic food webs. The study, published (DOI: 10.1007/s11783-026-2275-9) in the journal ENGINEERING Environment (Volume 20, Issue 11, 2026), analysed 498 organisms representing 34 species collected from Liaodong Bay (marine) and the Songhua River (freshwater) in northeastern China. The researchers found striking differences. In the marine food web, tri‑n‑propyl phosphate (TPP) dominated, with median concentrations of 147 ng/g lipid weight. In the freshwater system, tris(2‑chloroethyl) phosphate (TCEP) was the most abundant,...

I like the idea, not the propaganda. "CAP4 arylation is completed in 5 min versus 1 h for IAA alkylation". Nobody uses 1h IAA or CAA incubation and TCEP reduction can be done in 5 min, they are doing in 30 min. Nothing special to see here.

1. Data available upon reasonable request." It is an MS journal 🤦‍♂️.
2. 50-100 mM TCEP is the stock concentration. Sample prep is 10x lower.
3. Overnight incubation with 100 mM 🤔.
4. With some effort, we can produce any artifacts.
