Good question, we wondered too. For SEM the cells are dehydrated and imaged under high vacuum, and Methanosarcina has no rigid cell wall, just a thin protein S-layer, so they deflate a bit. But they aren’t perfect spheres anyway: in DIC (left) the surface is already uneven. Right: F420 autofluoresce

Need to pull a label out of green autofluorescence, or apart from a GFP sensor in the same channel? RSpFAST is a wash-free FAST label that light can also switch off: with little fluorogen and more light, the fluorogen leaves the pocket, then rebinds in the dark within seconds.

Geographic atrophy growth rates in the Amish population: findings from the Amish eye study
WHAT IS ALREADY KNOWN ON THIS TOPIC * Geographic atrophy (GA) secondary to age-related macular degeneration enlarges progressively over time and is a major cause of irreversible central vision loss. * GA growth rates vary substantially between eyes and are influenced by baseline lesion characteristics, including lesion size, focality, foveal involvement, fundus autofluorescence (FAF) phenotype and optical coherence tomography (OCT) biomarkers. * The Amish population is a genetically homogeneous founder population with relatively uniform lifestyle and environmental exposures, but longitudinal GA enlargement in this population has not been well characterised.WHAT THIS STUDY ADDS * This study provides 2 year natural-history data on GA enlargement in the Amish population using fundus autofluorescence imaging from the Amish Eye Study. * GA lesions in this cohort enlarged significantly over 2 years, with a mean annual growth rate of 1.35 mm²/year and a square-root–transformed growth rate of approximately 0.25 mm/year. * Baseline foveal involvement was the strongest predictor of subsequent GA enlargement and remained the only...

Targetoid Maculopathy in CEP290-Related Joubert Syndrome
An 8-month-old female infant, referred from pediatric neurology for pendular nystagmus and motor delay, exhibited ptosis, exotropia, and high hyperopia (cycloplegic retinoscopy: +8.50 + 1.50 × 90° right eye; +10.50 + 1.50 × 90° left eye). Ophthalmoscopy revealed myelinated retinal nerve fibers surrounding the right optic disc, alongside subtle epiretinal membrane and retinal pigment epithelium alterations in both eyes. Fundus autofluorescence depicted bilateral targetoid macular rings (A, B). Brain magnetic resonance imaging demonstrated the classic “molar tooth sign” (C) and a batwing-shaped fourth ventricle (D).

Presumed Pembrolizumab Maculopathy in a Woman with Ovarian Cancer
A 53-year-old woman with metastatic ovarian cancer receiving pembrolizumab presented with a generalized skin rash and blurred right-eye vision. Best-corrected visual acuity was 20/40 (right eye [OD]) and 20/20 (left eye [OS]). Fundoscopic examination was unremarkable (A), but fundus autofluorescence showed mild hyperautofluorescence around the fovea (B). OCT demonstrated attenuation of ellipsoid and interdigitation zones with relatively intact external limiting membrane and retinal pigment epithelium (arrows) in both eyes (C).

McArdle Disease
A 61-year-old visually asymptomatic man with muscle biopsy-confirmed McArdle disease had multiple reticular pigmentary changes in the macula and nasal midperiphery (A). Spectral-domain OCT demonstrated retinal pigment epithelium irregularities with focal disruption of the overlying ellipsoid zone (B). The radiating lesions were hypoautofluorescent and hyperautofluorescent with patches of atrophy on autofluorescence imaging (C1, C2). This patient has an extensive form of the pattern dystrophy extending beyond the macula compared with what has been previously described.

Anterior Segment OCT in Bilateral Diffuse Uveal Melanocytic Proliferation
A 60-year-old man with metastatic renal cell carcinoma previously treated with nivolumab presented with counting-fingers vision in both eyes. A, Fundus autofluorescence shows hyperautofluorescent and hypoautofluorescent changes. B, OCT shows retinal pigment epithelium undulation, subretinal fluid, and peripapillary bacillary layer detachment. C, Slit-lamp photography of the left eye shows anterior capsular pigment deposition. D, Anterior segment OCT of left eye demonstrates ciliary body thickening (red arrows) and hyperreflective deposits along the corneal endothelium (green arrows) and anterior lens capsule (yellow arrows).

Sunburst Chorioretinal Folds in Macular Neovascularization
A woman in her 70s with neovascular age-related macular degeneration in the fellow eye had her right eye imaged for 5 years with no abnormalities (A, 2018). The eye developed neovascular age-related macular degeneration with a large central pigment epithelial detachment (B, 2023). After 3 monthly intravitreal bevacizumab injections, as the pigment epithelial detachment contracted, radial chorioretinal folds resembling a sunburst appeared on autofluorescence imaging (C, 2024). The hyperautofluorescent lines reflect crowding of retinal pigment epithelium pigment as the retinal pigment epithelium–Bruch membrane complex is thrown into folds.
