We next treated migrating T cells (no targets present) with thapsigargin to pharmacologically deplete ER calcium stores. Surprisingly, this was sufficient to drive organelle trafficking to the centrosome but not polarisation of the multi-organelle ensemble to the plasma membrane.

We coin the phenomenon of Centrosome Organised Multi-organelle Ensemble Trafficking as COMET. We are now trying to determine how this process is regulated molecularly, whether it is shared in other immune cells and the role of organelle & their contacts in T cell killing.

Model: after target recognition synaptic actin is depleted, the perinuclear ER polarises leading to an increase in early ER-PM contacts and ER stores are partially emptied. This drives organelle trafficking to the centrosome, and the multi-organelle ensemble then polarises to the synapse.

Leveraging our advances in multispectral live imaging we find a characteristic three-phase program of polarity: i) the perinuclear ER first polarises, ii) organelles then cluster around the centrosome at the cell rear and iii) this multi-organelle ensemble polarises as a single unit to the synapse.

Proud of our work led by Andrew Mead in Nature Communications on apicoplast segregation in T. gondii. Also a wonderful co-submission with the Gubbels lab - together telling a great story of this new mechanism of organelle partitioning.
