Cryo-EM structure of a methanogen nitrogenase–PII protein supercomplex - Nature
Cryo-electron microscopy of a nitrogenase supercomplex from a methane-producing archaeon reveals a mechanism in which PII proteins drive assembly of such structures to regulate nitrogenase activity in response to nitrogen availability.

1/5 Cryo-EM gives high-resolution RNA structures, but only when high Mg²⁺ freezes out their motion.

At physiological conditions, RNAs are dynamic and 3D reconstruction fails. Can we still learn their folding equilibria?

New preprint: yes, straight from the raw particle images👇

Using cryo-EM, we showed that RNF213 forms a zinc-dependent 1.2 MDa dimer, occluding the E2 binding site and catalytic RZ domain to keep the E3 ligase inactive.

Cryo-tides, and now cryoSHIELDs. Maybe we need to get a peptide synthesiser to the EM facility?
Our latest development is now on bioRxiv: cryoSHIELDs — amphipathic peptide additives that make cryo-EM grid prep far more reliable. Tested across 24 protein systems, they enabled the first ABCC6 structure and GPCR–G protein structures at just 0.1–0.5 mg/mL!

We found that some of these mutations rearrange RNF213 to prevent dimer-mediated inhibition. Cryo-EM, biochemical assays and cellular investigation revealed that some mutants can even achieve the active conformation in the absence of ATP, resulting in increased inflammatory signaling.
