#micronuclei
Instead of manually counting and drawing boundaries around micronuclei, tiny DNA-containing structures that signal genome instability, we developed an AI-based tool that automates the process and helps researchers study how cells accumulate genetic damage.

We purified full-length human CIP2A and found remarkably long structures: CIP2A rods that form via a short tetrameric antiparallel coiled coil. The tetramerization is crucial for POLQ-mediated signatures of mitotic DNA repair. Interfering with CIP2A tetramerization hypersensitizes HR-deficient cells

☕Lebrec, @zierlab.bsky.social & co present a FRET-based cGAS reporter and show that cGAS activation by genotoxic stress is rare and uncoupled from micronuclei. The study implicates R-loop structures and subtypes of chromatin bridges as potential cGAS activators.

A fascinating study that sheds new light on the contribution of the cGAS-STING pathway to innate immune signalling following DNA damage.

A must-read for anyone working at the intersection of DDR and immune signalling in cancer research! 🧬🔥

Huge congrats to all the authors on this excellent work!

Applying this to DNA damage, we provide further evidence to work by @boultonlab.bsky.social, @makotothayashi.bsky.social and @acebronsp.bsky.social that cGAS enrichment on micronuclei does not generally lead to its activation, and we show that this is also not required for activation.

With an updated genetic cGAMP reporter, we reveal that cGAS activation by DNA damage is a rare event, and that cGAS enrichment on intracellular structures cannot be taken as evidence of activation.

This function of the S protein required its cleavage by proteases at the S1/S2 & the S2' sites. cell fusion damaged nuclei and resulted in the formation of micronuclei that were sensed by the cytosolic DNA sensor cGAS and led to the activation of its downstream effector STING.
