In every vertebrate studied to date, neuromesodermal progenitors (NMPs) — the bipotent stem cells that sustain axis segmentation — exist as a single tailbud pool.

In eels, we discover TWO spatially distinct NMP pools.

Microsurgical amputations show eels segment even without a tailbud !

Second: Germ layer fate restriction begins before gastrulation and extends beyond it. Detectable germ layer bias in the epiblast first appears ~E4. By contrast, in the growing tailbud bipotent axial progenitors continue to specify additional mesoderm and ectoderm through E9.5

The goal is to turn on Mesp in S-I and activate Notch in posterior S0. Tailbud elongation pulls FGF down, and so now we have NOT FGF AND Tbx6 satisfied in S-I! All that is needed is Notch! (18/n)

Posteriorly, there is a source of FGF that diffuses upward, but tailbud elongation pushes the source down over time. To get Mesp expression, you need Notch AND a protein called Tbx6 but NOT FGF. The segmentation of the somites involves creating this combination. (16/n)

Segments are normal sized, so is overall segment number and axis length (when tailbud is amputated).

I do think this is an eel-specific feature since the distribution of NMPs is quite unique and the pools are utilized differently over somitogensis.
