What is the connection between proteostasis and longevity?

This is one of the big questions my lab is currently tackling. We're looking for a #postdoc to take ownership of this project and help uncover novel regulators of proteostasis and aging.

🔬GPCRs are linked to many chronic diseases, acting as a central regulator of health. Therapies that rebalance GPCR signaling across tissues could enable a broader therapeutic effect that could potentially extend healthspan and prevent age-related diseases.

Impact of Chatbot-Based Cognitive Training on Cognitive Function and White Matter Integrity in Cognitively Unimpaired Older Adults: Randomized Controlled Trial
Background: Cognitive training may enhance cognition and support brain health during aging, but few trials have evaluated readily accessible chatbot-based programs with neuroimaging outcomes. Objective: This study aimed to evaluate the effects of a chatbot-based cognitive training program on white matter microstructural organization and cognition in cognitively unimpaired older adults. Methods: In a single-blind randomized controlled trial, 72 cognitively unimpaired older adults were assigned to chatbot-based cognitive training (30 min/day, 5 days/week for 12 weeks; n=37) or a no-intervention control (n=35). Diffusion tensor imaging at baseline and 12 weeks quantified fractional anisotropy (FA) in predefined white matter tracts, and cognition was assessed with the Cambridge Neuropsychological Test Automated Battery. Results: Groups were comparable at baseline. After 12 weeks, the training group showed significantly less FA reduction than controls in the bilateral tapetum and right posterior thalamic radiation, surviving false discovery rate control (P

Rob McConnell Interviews - BEN ABBA - Secrets of an Immortal
What if someone told you that a man is alive today who has witnessed nearly 2,800 years of human history? What if this individual had lived through the rise and fall of empires, the birth of major religions, devastating wars, remarkable scientific discoveries and countless generations of human civilization? And what if he claimed to possess the secret to living not merely 100 or 150 years—but potentially forever?Join Rob McConnell on The 'X' Zone Radio/TV Show as he welcomes Ben Abba, author, researcher, security professional and self-described investigator of human longevity and physical immortality, for an extraordinary exploration of one of humanity's oldest and most tantalizing mysteries: Can human beings overcome aging and death? For thousands of years, humanity has searched for the secret of eternal life. From the legendary Fountain of Youth and the ancient alchemists' quest for the Elixir of Life to modern genetic research and regenerative medicine, the desire to extend human existence has fascinated philosophers, scientists, spiritual teachers and explorers. But Ben Abba believes that the answer may already exist—and that extraordinary individuals possessing the secret to exceptional longevity may be living among us.Drawing upon his background in computer security, financial investigations and research, Abba set out to locate individuals who had allegedly lived far beyond the limits of ordinary human life.Initially, his objective was relatively straightforward: find the oldest person alive and discover what could be learned from that individual's lifestyle, habits and personal history.According to Abba, his investigation led him to people claiming extraordinary ages, including individuals he believed had lived for more than 150 years. But the most astonishing development came when he reported making contact with a man whose claimed age approached 2,800 years.Abba maintains that this individual had personally witnessed events spanning much of recorded Western civilization.Imagine speaking with someone who claimed to have lived during the age of ancient Greece, witnessed the emergence of the Roman Empire, observed the development of Christianity and survived through the Middle Ages, Renaissance, Industrial Revolution and modern technological age. What stories could such a person tell? What historical events might they remember differently from the accounts preserved in textbooks? And what could they reveal about the human experience after centuries—or millennia—of existence? These extraordinary assertions became the foundation of Abba's project, Secrets of an Immortal: An Eyewitness Account of 2,800 Years of History.Through his research and interviews, Abba explores the possibility that certain individuals may have discovered ways to dramatically slow the aging process or extend their physical lives beyond currently documented human limits. He has also discussed his belief that approximately two dozen people with extraordinary longevity may be living in the world. His investigations extend into subjects including consciousness, spiritual development, reincarnation, remote viewing, personal belief and the relationship between the mind and physical health.Abba has described how his search for extraordinary longevity changed his own outlook on mortality and the possibilities of human existence. He suggests that fear of death can influence the way people live and that reconsidering assumptions about aging may encourage individuals to approach life with greater curiosity and purpose. However, the central claims raise substantial questions. How could someone prove they were nearly 2,800 years old?Would historical records, biological samples, genetic analysis or independent documentation be sufficient to verify such an extraordinary lifespan?Could a person's detailed knowledge of ancient history establish that they personally witnessed those events? And why would someone who had allegedly survived for thousands of years choose to remain unknown to the wider world? These are precisely the kinds of questions that make Abba's story so provocative.Modern science has documented exceptional human longevity, with Jeanne Calment of France reaching a verified age of 122 years and 164 days. Researchers continue studying the biological mechanisms of aging, but there is no independently verified evidence that any human being has lived for centuries, much less thousands of years.Claims of extreme longevity require rigorous examination of birth records, identity, historical continuity and biological evidence.Likewise, while healthy habits can improve well-being and reduce certain health risks, no scientifically established practice can make a human physically immortal.Nevertheless, the desire to understand aging remains one of the most important subjects in medicine, biology and philosophy.

Detection of Age-Related Cognitive Decline Over Short Time Frames Using Digital Multiday Learning Assessments: Longitudinal Study
Background: High-resolution digital cognitive tools administered at frequent intervals enable the study of age-related cognitive change at a previously unattainable level of granularity. Computational modeling further enables the simultaneous examination of multiple aspects of cognitive performance. Objective: The objective of this study was to examine whether repeated digital multiday learning assessments can detect age-related cognitive decline over short time frames in cognitively unimpaired older adults and to quantify age-related differences in process-based cognitive markers derived from computational modeling. Methods: A total of 209 cognitively unimpaired older adults (mean age 73.6, SD 8.2 years; n=136, 65.1% female) from the Harvard Aging Brain Study and affiliated studies were administered the Boston Remote Assessment for Neurocognitive Health, which includes the face-name task, a paired associative learning task. Daily median response times for correct responses were used as the primary outcome. A new version of the face-name task was administered across up to 4 testing intervals over a mean of 2.8 (SD 0.4) years. Using computational modeling, we estimated longitudinal changes in peak performance, a practice-adjusted marker of an individual’s best expected response time, as an indicator of age-related cognitive decline. We further examined age-related differences in peak performance, stability (within-person variability), learning within and across testing intervals, and between-interval forgetting, with embedded linear regressions for age, sex, years of education, and device type. Results: Peak performance slowed by 0.04 seconds per year (95% credible interval 0.02-0.06; approximately 2% per year), indicating age-related cognitive decline. Higher age (+1-SD) was associated with slower peak performance (+0.19 seconds, 95% credible interval 0.12-0.26), steeper annual slowing in peak performance (+0.036 seconds per year), greater within-person variability (+0.043 seconds), reduced learning across testing intervals, and more forgetting. Conclusions: Our findings indicate that a digital multiday learning paradigm combined with computational modeling of response times sensitively detects age-related cognitive decline over short timescales (
