This week in #Science #Immunology: Peng Shao, Noah Butler, & colleagues at University of Iowa show that TRAF3 is critical for early #TFollicularHelper ( #Tfh ) cell differentiation!
TRAF3 is critical for initial T follicular helper cell specification via coordination of the IL-6R/IL-2R–BCL6 signaling nexus
TRAF3 integrates early cell-intrinsic and cell-extrinsic signals to initiate TFH cell differentiation.

This work is also important as it reveals that like BCL6, SIPR2 is expressed on activated B cells prior to GC entry and marks cells that will go on to become optimally responsive IgM+ MBCs. It also reveals that CD4-Cre BCL6-floxed CD4+ T cells are still capable of migrating to the T-B border. ⤵️

I am certain there is a really good evolutionary reason probably related to autoimmune disease that BLIMP1 and BCL6 are mutually repressive but it would be super cool if we could make them… not. In some circumstances.

My favorite thing about T cell biology is how it’s so tractable and finite. Anyway, here’s a preprint showing that there are TFH cells that don’t express BCL6, express BLIMP1, and they seem to be important for plasma cells:

The project started when Zosia was a summer student, & we discussed the great paper by Słabicki et al. The ubiquitylation of BCL6 by SIAH1 is boosted when BCL6 gets multimerised by a small molecule. We reasoned that SIAH1 might also be a multimer & preferentially recognise multimers through avidity.
