Pairing this assay with our cichlid GCaMP calcium imaging will let us watch neural activity while they perform the behavior—connecting sensory processing, motor control, and evolutionary change in a highly accessible vertebrate system. Looking forward to more #neuroethology in this model!

2 beautiful cells expressing #GCaMP in #chicken retina via AAV-mediated retinal #transduction.
The dendritic fields of these cells are vast, beautiful and ordered, depicting how truly intricate #retina connectivity is. #Avian retina is remarkable.

My choice for today's #FluorescenceFriday image.

With everything that's going on, I needed this cool data 1 day before #flourescencefriday. It's my first time with #GCaMP imaging (green), and you can clearly see Aβ42 induced hyperexcitability at the same time that we also see BBB leakage (not shown)! #zebrafishrock

Completely agree! E.g: In this preprint, I co-expressed axon-GCaMP and inhibitory DREADD (H4) and showed that local CNO infusion attenuates terminal Ca2+ during singing. In my case, I wanted to validate the DREADD, but this can also be used the other way around (1/3)

How we did it: we register 4 views together, in-vivo 2p GCaMP functional, in-vivo GCaMP z-stack, ex-vivo GCaMP confocal and finally MERSCOPE imaging from @vizgen_inc.

fluorescence lifetime of GCaMP, i.e. how long it takes GCaMP to emit a photon after excitation. In this paper, they make a GCaMP6s variant that is ~3x faster than the original using a fluorescent protein called BrUSLEE. This makes sense because Bruce Lee was ~3x faster than a normal human 3/

Check out this compact 920 nm ultrafast fs laser for 2p imaging of GCaMP, etc.. Mounted on its side like this, it takes up very little space on the table.

That's the Axon 920, generously loaned to the course thanks to @ewadorotaz.bsky.social and others at Coherent. It's what we use on our own rigs.

A basic image for #fluorescencefriday -- neurons expressing the Ca++ sensor, GCaMP, imaged with a 2-photon microscope. The cool thing is the microscope is built by neuroscience students on our PhD optics course. It is the last thing they build at the end of their two week optics journey.

#FluorescenceFriday rehash. DRG cultures expressing ER-targeted GCaMP and treated with CMXRos. May be “old” data, but finding it tonight instantly reminded me of how much I love these organelles.

To test for crosstalk, we compared activity in overlapping GCaMP and dTomato signals. Calcium events in the GCaMP channel showed no corresponding change in dTomato, confirming minimal crosstalk between channels 🎯
