AAN-I-IFNs in COVID-19 pts linked to 🔺nasopharyngeal viral load (P=1.4x10⁻³) & delayed clearance (P=.013). These + high viral load = risk factors for critical disease.⚠️
High Nasopharyngeal SARS-CoV-2 Load and Delayed Clearance in Hospitalized Patients With Blood Autoantibodies Neutralizing Type I Interferons
AbstractAutoantibodies neutralizing type I interferon (AAN-I-IFNs) underlie life-threatening COVID-19. In hospitalized COVID-19 patients of the DisCoVeRy trial (NCT-04315948), AAN-I-IFNs were associated with higher nasopharyngeal viral load at inclusion (P = 1.4 × 10−3) and delayed viral clearance (P = .013). Blood AAN-I-IFNs, high viral load and delayed clearance were also independent risk factors for critical COVID-19.

🧨Interferon Production: Upstream kinases like MST1/2 can directly enhance the activation of IRF3 (Interferon Regulatory Factor 3). IRF3 then enters the nucleus to trigger the production of Type I Interferons (IFNs), which are the body's premier first-line defense chemicals against viruses

Since TBK1/IKKe also function downstream of viral sensors to induce type I IFNs, this is likely an evolutionary adaptation to infection by pathogens that encode TBK1/IKKe antagonists. This would enable the infected cells to be eliminated by IFNg secreted by T and NK cells.
