Excited to share our new preprint! 📜

Here, we used CALVADOS and ultra-CG models to show how the kinetochore establishes a robust attachment to the dynamic microtubule end.

🔬 From JCB’s July issue (rupress.org/jcb/issue/22...): a PtK2 cell plated on a micropattern and injected with low doses of soluble actin (orange) and tubulin (blue) in order to generate speckles along the corresponding filaments. From @manuelthery.bsky.social group (rupress.org/jcb/article/...).

We detected specialized microtubule organizations that provided answers.
Early (left), microtubules formed a meshwork which was interwoven with Buc condensates.
Later (right), microtubules formed a cage-like organization that encapsulated the mature Bb.

We employ all-atom replica exchange molecular dynamics simulations to resolve a high-confidence neck-region conformation of the dimeric human kinesin-1 bound to a structurally realistic microtubule lattice and use this structure to simulate kinesin’s initial stepping motion.

This surface pattern reflects a patterned cytoskeleton. Actually Stentor has two cytoskeletons: microtubules form parallel bundles called KM fibers (left), while centrin forms a network of contractile fibers called myonemes (right).

I've always preferred cyan, yellow, and magenta. Magenta and green work well, but cyan and green are too similar. Yellow, cyan, and magenta work well too. Example: EM of actin filaments (magenta), microtubules (immunogold, yellow), and clathrin pits (cyan).
