SOD1 is one such gene, where toxic gain of function variants cause the SOD1 protein to misfold and aggregate, which leads to motorneuron death. Tofersen causes targeted cleavage of SOD1 transcripts (mutated and wild type), resulting in less SOD1 production and less misfolding.
