Biotic interactions shape the realized niche of toxic cyanobacteria | Nature Ecology & Evolution
Lake Greifensee is periodically affected by cyanobacterial blooms and associated cyanotoxins20,21,22 (Fig. 1). Our study focuses on two key groups: the toxic Microcystaceae group (Microcystis spp., Aphanothece spp., Aphanocapsa spp.) and Dolichospermum spp. Microcystacae are the most common potentially toxic bloom-forming cyanobacteria worldwide23, typically blooming in Greifensee during summer and autumn (Fig. 1c). Previous studies confirm the production of microcystins and numerous other bioactive metabolites from this group in the lake, including during our study period20,22. Dolichospermum, the second most common potentially toxic bloom-forming cyanobacterium globally, is known to produce both hepatotoxins and neurotoxins3,24. While rarely forming surface blooms in Greifensee, it reaches peak abundance in late summer, and bioactive metabolites potentially associated with this taxon are found during the study period22. Our high-resolution data reveal significant temporal fluctuations in both cyanobacteria (Fig. 1c). This provides a unique opportunity to examine growth dynamics and their relationships with biotic and abiotic factors at temporal scales directly relevant to bloom formation. To quantify these dynamics and investigate the influence of biotic interactions, we present an nGR analysis based on high-frequency monitoring data. Quantification of bioactive compounds in the juices of 21 American elderberry cultivars
American elderberry (Sambucus nigra subsp. canadensis) is recognized as a rich source of bioactive compounds with nutritional and therapeutic potential. However, comprehensive quantification of its metabolites remains analytically challenging due to the structural complexity of phenolics and matrix interferences in juice extracts. In this study, a targeted ultra-high performance liquid chromatography-tandem mass spectrometry (UHPLC-MS/MS) workflow was developed and validated to quantify 22 bioactive metabolites, including anthocyanins, flavonoids, and phenolic acids, in juices from 21 American elderberry cultivars. An optimized UHPLC-MS/MS system demonstrated higher sensitivity, selectivity, and throughput than conventional high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), achieving limits of detection and quantification below 0.1 ng/mL for most analytes. Matrix effects were effectively mitigated through sample dilution, enabling accurate and reproducible quantification across complex juice matrices. Quantification of metabolites showed that accession 1199 exhibited high levels of cyanidin-based anthocyanins, suggesting its utility as a natural food color source. Overall, high-throughput UHPLC-MS/MS method provides a robust analytical framework for metabolomic profiling for cultivar selection and the development of elderberry-derived functional products.

The global effort to eradicate malaria requires the development of drugs with novel mechanisms of action and differentiated pharmacological profiles. Both liver and blood phases of Plasmodium infection are essential for disease progression and transmission, making them primary targets for therapeutic intervention. In this study, we established a dual-luciferase reporter assay to simultaneously assess Plasmodium berghei liver-stage development and host cell viability. Screening a library of bioactive small molecules with annotated human targets revealed several inhibitors with potent liver-stage activity and minimal cytotoxicity. Further profiling against P. falciparum asexual blood stages identified compounds with dual-stage activity, underscoring their potential utility for development as either prophylactic or therapeutic agents. Knowledge of their human targets will facilitate investigation into host- or parasite-directed antimalarial mechanisms, thereby accelerating both biological insight and rational drug development. Our findings constitute a valuable resource for research on malaria biology and the discovery of next-generation antimalarials.
