Lung tumors mis-splice STAT2, a key immune gene, building a broken form that actively jams the signal rather than staying quiet. That splicing glitch associates with immunotherapy response. 🧵

Alcohol-Related Liver Disease Linked to RNA Splicing Errors and Inflammation
A study published in Nature Communications reveals that alcohol-related liver disease traps damaged cells in a regenerative limbo, preventing recovery even after abstinence. Researchers from the University of Illinois Urbana-Champaign, Duke University, and the Chan Zuckerberg Biohub Chicago discovered that inflammation disrupts RNA splicing, causing cells to remain stuck between mature and progenitor states. This cellular dysfunction leads to liver failure as healthy cells overwork and fail to regenerate properly. The study highlights a key pathway involving ESRP2 protein deficiency and inflammatory signals, suggesting potential treatments targeting inflammation could restore liver repair. Findings may lead to diagnostic markers for alcohol-associated liver disease and therapies addressing RNA splicing defects. This research addresses the global issue of alcohol-induced liver mortality, which causes approximately 3 million deaths annually.

The fix: a #minigene assay. Synthetic DNA spanning the variant is cloned into a #splicing reporter, expressed in cells, then read out by RT-PCR & Sanger sequencing — no patient #RNA needed. Turnaround: ~9-10 days, fast enough for clinical use.

Poly(A) selection limits detection of long and alternatively spliced transcripts compared with rRNA depletion in RNA-Sequencing
The eukaryotic transcriptome diversity arises largely from alternative splicing. One of the widely used high-throughput methods to study this diversity is RNA sequencing. RNA sequencing has become a c...
