CRIF1 Linked to Aggressive Endometrial Cancer and Reduced Response to Paclitaxel Treatment
A recent study published in Scientific Reports has identified CRIF1 (CR6-interacting factor 1) as a key factor in promoting aggressive phenotypes in endometrial cancer (EC) cells and reducing the effectiveness of paclitaxel, a common chemotherapy drug. The research, conducted by a team from Shanghai Pudong Hospital of Fudan University, examined 95 endometrial cancer tissues and matched adjacent normal tissues using immunohistochemistry, RT-qPCR, and western blotting. The findings revealed that CRIF1 expression was significantly upregulated in EC tissues compared to normal tissues, with higher expression levels correlating with advanced FIGO stage, higher histological grade, deeper myometrial invasion, and increased Ki-67 positivity. Under baseline conditions without chemotherapy, CRIF1 overexpression increased the proliferation and migration of Ishikawa cells (a common endometrial cancer cell line). Following paclitaxel treatment, CRIF1 overexpression was associated with reduced apoptosis, lower oxidative stress, enhanced glycolytic activity, and increased Wnt/β-catenin-related signaling. The study also investigated the role of Wnt/β-catenin signaling using Wnt-C59, a specific inhibitor, which partially attenuated the effects of CRIF1 overexpression, suggesting that this signaling pathway plays a significant role in the observed phenomena. The researchers concluded that CRIF1 may have context-dependent effects in endometrial cancer and could be associated with cellular adaptation to paclitaxel-induced stress. While the study provides important insights into the mechanisms of chemoresistance in endometrial cancer, it also highlights that further research is needed to determine whether CRIF1 influences responses to other chemotherapeutic agents or contributes to clinically relevant paclitaxel resistance. This research has significant implications for understanding the molecular mechanisms underlying endometrial cancer progression and treatment resistance, potentially leading to new therapeutic strategies targeting CRIF1 or the Wnt/β-catenin pathway to improve patient outcomes.
