Microglial TDP-43 loss affects myelin and Tyrobp regulation in mice. Changes observed via MRI, confocal, and EM imaging. 🧠🔬 PMID:42420559, Nat Neurosci 2026, @NatureNeuro https://doi.org/10.1038/s41593-026-02348-3 #Medsky #Pharmsky #RNA #ASHG #ESHG 🧪
Microglial TDP-43 mediates myelin refinement and represses Tyrobp cryptic exon inclusion in mice | Nature Neuroscience
TDP-43 proteinopathy is a hallmark of neurodegenerative disorders such as amyotrophic lateral sclerosis and frontotemporal dementia where mislocalization of TDP-43 has been observed in neurons and glial cells. However, the role of TDP-43 in microglia and the consequences of its loss of function remain unexplored. Combining magnetic resonance imaging, and confocal, and electron microscopy, we uncovered structural changes and myelin abnormalities in the early postnatal brain of mice lacking microglial TDP-43. Spatial transcriptomics further revealed an enriched interferon-responsive signature associated with oligodendrocyte dysfunction. Early depletion of microglial TDP-43 led to motor deficits in adult mice. Mechanistically, knocking out TDP-43 impaired microglial ability to engulf and degrade myelin. It also led to cryptic exon inclusion in the Tyrobp mRNA, resulting in truncated DAP12 protein, thus causing defective TREM2 signaling. Our findings reveal a role for TDP-43 in regulating

5/ The result is a fly whose eyes are built on the butterfly plan, 9 cells per unit instead of 8, with the same three-way random mosaic butterflies have. We've been calling it the butterfly-fly.

6/ The brain didn't need to change at all.

Fly brains overbuild a class of partner neurons, and the extras die when they find nothing to connect to. Given something to connect to, they lived, and wired to the correct subtype on the first try, with no further genetic change.

4/ Butterflies changed the identity of those two neighbors, so they never close ranks. The mystery cell stays, keeps contact with the center, and becomes a second R7.

We found the gene, knocked it out in butterflies, then put the change into a fly.

7/ There's a hawkmoth that looks like it's partway through the same transition: the lower half of its eye is butterfly-like with two of these cells, the upper half is fly-like with one. Roughly what you'd expect if the change started in one region and spread.

Most blue coloration in animals is structural, not pigments & relies on biogenic guanine crystals.

Guanine is the "G" base in your DNA.

When crystallized in a living cell, it forms thin layers of high refractive index like a thin mirror. Stack these up & you get prismatic color.
