Online now: Notch3 destabilizes regulatory T cells to drive autoimmune neuroinflammation in multiple sclerosis
Notch3 destabilizes regulatory T cells to drive autoimmune neuroinflammation in multiple sclerosis
Benamar et al. demonstrate that Notch3+ Treg cells are increased in multiple sclerosis and in EAE. They are induced in the gut by microbiota and translocate to the CNS to degenerate into pathogenic Th17 cells. They also outcompete a protective NPY1R+-resident Treg cell population that controls Th1 responses.

Boston Children’s Hospital and collaborators studied patients with multiple sclerosis and mice.

They found that a gut-driven subset of Treg cells carrying Notch3 travel to the brain, lose their calming role, and turn into inflammatory Th17 cells.

Multi-omic profiling reveals pericyte and smooth muscle cell contributions to CADASIL pathology in cell-specific Notch3 mutant mice
Huang et al. identify cell-type-specific mechanisms driving CADASIL pathology using conditional Notch3R170C mouse models. Pericyte and smooth muscle cell mutations produce distinct vascular changes, inflammation, and memory deficits. Multi-omic profiling reveals mural cell-specific pathways that contribute to the CADASIL-associated pathology.
