To supplement our cryo-ET and generate candidates for proteins that localize to different ciliary regions, we took a trip to the local butcher to pick up some cow trachea to build a mammalian multiciliated cell interactome using XL/MS (enjoy this vid of @cellarchlab.com shaking cilia off trachea)

LLOMe has long been used to study lysosomal damage, yet how it works has remained a mystery.
Using cryo-electron tomography, we show it forms amyloid structures inside lysosomes that mechanically rupture membranes – revealing a new paradigm for lysosomal failure.

Our recent eLife paper is now an official version of record. This was my first project using #Cryo-ET and I'm definitely hooked - it's so fun to look at snapshots of synapses within brain tissue and see individual proteins. Excited to share more soon!

First time posting on Bluesky! 🥳 We discovered a new pilus family in cyanobacteria. The pilin has a novel protein fold that forms a super-tight double-layer tube. Similar gene clusters exist in a few cyanobacterial orders. A great collaboration with John and others! www.nature.com/articles/s41...

Proudly present our new preprint on my first post here. We performed SPA on the ciliary tip and identify 7 proteins. Among them, we found the conserved protein SPEF1 at the microtubule seam. In vitro assays show that SPEF1 binds to the seam and stablizes microtubules.

🎉 Out today in Cell: the paper I've been looking forward to sharing for a long time 🎉

We used cryo-ET to find a molecular machine nobody knew existed on the surface of a minimal bacterium, and worked out what it does.
